- [LSM700 Upgrade Announcement]
Attention: As the LSM700 parts have been discontinued, to avoid months of temporary downtime during future failures, the center will be upgrading the equipment from late September to early October (one week in advance), so please plan and arrange your experiments in advance to minimize the impact of downtime.
After the upgrade, the quality and speed of image acquisition will be improved. However, the acquisition software, scan area, intensity, etc., will differ (it cannot be applied to the original LSM700 settings), and the original users must be re-qualified to use the upgraded model. The center will arrange the training course and assist in re-qualifying.
Microscope:
Image Software:
Note 1. LSM700 Scan Head:
LSM700 is equipped with two sets of PMTs and a VSD Beam Splitter to provide signal acquisition band flexibility with an adjustable accuracy of 1 nm. It is not limited by traditional filters and can collect more signals.
- Fully motorized inverted microscope Axio Observer Z.1
- Electric XY platform can be used for large-area map images and multi-position imaging
Objectives:
- Fluar 5x/0.25
- Fluar 10x/0.5 DIC
- Plan-Apochromat 20x/0.8 DIC
- Plan-Apochromat 40x/1.4 Oil DIC
- Plan-Apochromat 63x/1.4 Oil DIC
- Plan-Apochromat 100x/1.40 Oil DIC
Laser (nm):
- Solid-state laser: 405nm, 488nm, 555nm, 639nm
Detectors:
- Two sets of high-sensitivity PMT with fixed fluorescent filters and adjustable VSD BeamSplitter (Note 1).
- A set of T-PMT dedicated detector for high-sensitivity transmitted light images, including automatic switching light path control, which can acquire various transmitted light images, such as bright field and differential interference contrast (DIC) images.
Live-cell Incubation:
Stage-top live-cell incubation system with temperature, humidity and CO2 control module
The picture above shows [Stage-top live-cell incubation system PM S1], which is directly installed on the stage. The carbon dioxide concentration is stably controlled from 0.0% to 10.0% (±0.1%). It is suitable for glass-bottom dishes with diameters of 30-60mm and coverslip thickness of 0.17mm. If you need to use the live cell system, please contact the administrator before making your first appointment.
Image Software:
- Zen 2009, Confocal image acquisition and analysis ( LSM700 Operation Manual )
- Offline computer workstations are available for image processing and analysis
