Laser (nm):
Two-photon laser 690-1040 nm (Note1)

Detectors:
  • Two sets of reflective high sensitivity, low electronic noise photomultiplier sensors NDD (Non-Descanned Detectors), three sets of NDD detection wavelength ranges are SP485nm, BP500nm-550nm, BP565nm-610nm.
  • One set of T-PMT sensors for high sensitivity transmitted light images, with automatic switching of light path control, can capture various transmitted light images, such as bright field and differential interference DIC observation.
  • Built-in field aperture, light scattering plate, heat-absorbing/IR filter, switching light/infrared light to provide bright field, oblique illumination, and DIC (Differential interference contrast) contrast.

Note 1: Two-photon laser
  • Two-photon technology utilizes the energy of two double-wavelength photons to stimulate fluorescence in an additive manner.
  • Since the longer wavelength infrared light excites the sample and the two-photon is only imaged at the focal point, the sampling depth can be increased without pinhole filtering of the non-focal plane signals. It is beneficial for observing highly scattering samples, and 100% reflected fluorescence can be perceived.
  • In addition, the relatively low energy of infrared light can minimize photobleaching and cytotoxicity produced by photochemical reactions and is suitable for long-time live-cell image acquisition.

Users should pass the certification test after the instruction and operate the system more than five times (inclusive) during office hours (8:30-16:30) before they are authorized to reserve online, have access to Room 640 and can use the system according to the regulations. If the certified user does not use the device for one month, they must be certified again by the administrator.
  1. Reservation regulations:
    • Please go to the "Instrument Reservation System" to make a reservation before use. The person who makes the reservation must be the actual user.
    • After use, please register in the record book. The start time is the reservation time, and the end time is based on the end time of use. You cannot delay the start time or occupy other people's sessions.
    • Reservations cannot be canceled on the same day. Reservations made more than half an hour after the scheduled time are considered abandoned.
    • No same-day reservations are accepted. Please confirm your reservation by 16:30 of the previous workday at the latest; late changes violate the rules and will not be accepted.
    • Each person can make 4 reservations within two weeks, and each reservation can be made for 2 hours.
  2. Sample requirements:
    • Please use a fluorescent microscope to confirm the samples are fluorescent before loading. If the samples cannot be serviced due to their own factors, the center will still charge according to the hours of operation.
    • If you have special needs, please inform us of the dyes used for the samples before operations so the technician can understand and advise the user's needs.
    • Please contact the center administrator for any other needs or special requirements.

Remarks
  1. Abbreviations: (O) Olympus, (Z) Zeiss
  2. Number of Reservations: Each person can make up to four reservations on a single device within a two-week period. Nighttime use and LSM7MP allow up to 3 hours per reservation, while other devices allow up to 2 hours per reservation. Reservations can be made consecutively on all devices.
  3. The office hours are from 08:30 to 16:30, and the nighttime period is from 16:30 to 08:30 of the next day.
  4. If you need to take time-lapse imaging overnight, you must contact the administrator of the center in advance.
  5. LSM 7MP is open from 09:00 to 17:00. If you need to operate at night, you must inform the administrator in advance.
  6. In addition to the education and training of the imaging core facilities organized by the center, it is recommended that you prepare your own samples (experimental data can be taken away) for scheduled training and certification tests. The charges will be the same as for general use of the system.
  7. The above table shows the hourly usage charge. Charges are calculated on a half-hour basis.
  1. If you have the following qualifications, you will be charged the same fee as NYCU faculty and students:
    • Branch school
    • Joint-appointed faculty 
    • Affiliated hospitals 
    • Projects are done in cooperation with and under the control of the university.
  2. If you have the following qualifications, the fee will be the same as that of a cooperative educational unit:
    • Adjunct faculty
    • Teaching hospitals (Veterans General Hospital, Taipei City Hospital, Far Eastern Memorial Hospital, etc.)
    • University System of Taiwan (UST)
    • Companies participate in the academia-industry cooperation center of NYCU
Instruments are charged per half hour, and the billing will be counted at the end of each odd-numbered month. Please bring the bill to the Cashier's Section of the Administration Building within two weeks at the latest. The service will be stopped if the number of overdue payments reaches (and includes) two times.
  1. No USB flash drive is allowed. Please use FTP to transfer files or bring your blank CD-ROM to burn data. Due to the limited space of the computer's hard disk, the administrator will delete the data from time to time to facilitate the normal operation of the instrument. We are not responsible for data storage.
  2. Without the consent of the administrator of the Center, you are not allowed to add or remove any parts, alter the system settings, or install software on the Center's public equipment. If any experiment requires any test, the administrator's consent must be sought first and accompanied by the engineer of the original manufacturer to carry out the testing without damaging the normal operation of the equipment, and only after the passing of the test can it be operated and used by the personnel approved by the administrator of the Center under the condition of the administrator's permission.
  3. According to the administrator's judgment, the violator is warned for the first time, canceled the right to use for one month for the second time, and canceled the right to use permanently for the third time; if the violator violates the above specification, resulting in the damage of the instrument, the violator shall pay compensation to repair the instrument and stop the right to use for one month. The violator is prohibited from using the instrument permanently.
