  • Term of use: The specialist will help you operate the machine during working hours, and you can use it at night after certification.
    • In addition to the AiryScan function, it is open for self-operation certification.
    • Users should undergo training, pass the certification test, and operate the device five times (inclusive) independently during office hours. Then, they can book and operate it at night according to regulations.

  • The picture above shows [Stage-top live-cell incubation system PM S1], which is directly installed on the stage. The carbon dioxide concentration is stably controlled from 0.0% to 10.0% (±0.1%). It is suitable for glass-bottom dishes with diameters of 30-60mm and coverslip thickness of 0.17mm. 
  • If you want to use the living cell system, please call the administrator before making your first appointment.
  • With Definite Focus 2, an auto focus maintaining system.
Software:
  • Zen 3.2 (Blue) for imaging and analysis
  • An offline computer workstation is available for image processing and analysis

Note 1. The 32-channel AiryScan 2 matrix super-resolution detector offers 2Y and 4Y modes. The scanning speed in 4Y mode is 18.9 frames per second (512 x 512 pixels) or more, which is faster than its predecessor and results in a smaller file size.

AiryScan is ZEISS's latest imaging technology
While conventional confocal images utilize a pinhole aperture to filter light from the non-focal plane, AiryScan is the latest ZEISS imaging technology that uses a 32-channel planar detector to collect all the light from a single source simultaneously. Since each detector of AiryScan can be regarded as a tiny single pinhole aperture, it can exceed the optical limitations and improve image resolution and quality. Taking the 488nm excitation as an example, the resolution of the XY-axis can be increased to 140 nm and the Z-axis to 400 nm.

This function has yet to be opened for certification. If you need to use it, it will be operated by the center administrator.
Note 2.  To use the time-lapse live-cell imaging with the CCD camera, users must have been certified for confocal imaging and contact the administrator to make weekend reservations during office hours.

There is an operation service for LSM900 during office hours. It is also open for self-operation certification except for the AiryScan function.
For self-operation certification, users should pass the certification test after the instruction and operate the device independently more than five times (inclusive) during office hours (8:30-16:30). Certified users are then authorized to reserve and operate at nighttime according to the regulations. If the certified user does not use the device for one month, they must be certified again by the administrator.
  1. Reservation regulations:
    • Please visit the "Instrument Reservation System" to make a reservation before use. The person who makes the reservation must be the actual user.
    • After use, please register in the record book. The start time is the reservation time, and the end time is based on the end time of use. You cannot delay the start time or occupy other people's sessions.
    • Reservations cannot be canceled on the same day. Reservations made more than half an hour after the scheduled time are considered abandoned.
    • No same-day reservations are accepted. Please confirm your reservation by 16:30 of the previous workday; late changes violate the rules and will not be accepted.
    • Each person can make four reservations within two weeks, and each reservation can be made for 2 hours.
  2. Sample requirements:
    • Always use 0.17mm ( No.1.5) glass coverslips for slide samples or glass-bottom Petri dishes.
    • Please be sure to clean the surface of your samples before loading.
    • Please ensure the samples are sealed and that a fluorescent microscope is used to confirm the samples are fluorescent before loading. If the samples cannot be serviced due to their own factors, the center will still charge according to the hours of operation.
    • If you have special needs, please inform us of the dyes used for the samples before operations so the technician can understand and advise the user's needs.
    • Please contact the center administrator for any other needs or special requirements.

Remarks
  1. Abbreviations: (O) Olympus, (Z) Zeiss
  2. Number of Reservations: Each person can make up to four reservations on a single device within a two-week period. Nighttime use and LSM7MP allow up to 3 hours per reservation, while other devices allow up to 2 hours per reservation. Reservations can be made consecutively on all devices.
  3. The office hours are from 08:30 to 16:30, and the nighttime period is from 16:30 to 08:30 of the next day.
  4. If you need to take time-lapse imaging overnight, you must contact the administrator of the center in advance.
  5. LSM 7MP is open from 09:00 to 17:00. If you need to operate at night, you must inform the administrator in advance.
  6. In addition to the education and training of the imaging core facilities organized by the center, it is recommended that you prepare your own samples (experimental data can be taken away) for scheduled training and certification tests. The charges will be the same as for general use of the system.
  7. The above table shows the hourly usage charge. Charges are calculated on a half-hour basis.
  1. If you have the following qualifications, you will be charged the same fee as NYCU faculty and students:
    • Branch school
    • Joint-appointed faculty 
    • Affiliated hospitals 
    • Projects are done in cooperation with and under the control of the University.
  2. If you have the following qualifications, the fee will be the same as that of a cooperative educational unit:
    • Adjunct faculty
    • Teaching hospitals (Veterans General Hospital, Taipei City Hospital, Far Eastern Memorial Hospital, etc.)
    • University System of Taiwan (UST)
    • Companies participate in the academia-industry cooperation center of NYCU
Instruments are charged per half hour, and the billing will be counted at the end of each odd-numbered month. Please bring the bill to the Cashier's Section of the Administration Building within two weeks at the latest. The service will be stopped if the number of overdue payments reaches (and includes) two times.
  1. No USB flash drive is allowed. Please use FTP to transfer files or bring your blank CD-ROM to burn data. Due to the limited space of the computer's hard disk, the administrator will delete the data from time to time to facilitate the normal operation of the instrument. We are not responsible for data storage.
  2. Without the consent of the administrator of the Center, you are not allowed to add or remove any parts, alter the system settings, or install software on the Center's public equipment. If any experiment requires any test, the administrator's consent must be sought first and accompanied by the engineer of the original manufacturer to carry out the testing without damaging the normal operation of the equipment, and only after the passing of the test can it be operated and used by the personnel approved by the administrator of the Center under the condition of the administrator's permission.
  3. According to the administrator's judgment, the violator is warned for the first time, canceled the right to use for one month for the second time, and canceled the right to use permanently for the third time; if the violator violates the above specification, resulting in the damage of the instrument, the violator shall pay compensation to repair the instrument and stop the right to use for one month. The violator is prohibited from using the instrument permanently.
