A Spectrophotometer (spectrometer) is designed to detect wavelengths ranging from 190 to 900nm, encompassing both the ultraviolet and visible light spectrums. This instrument is primarily utilized in life sciences to measure the absorbance of samples within this range to determine the concentration of the sample. The accompanying software features two main measurement categories: nucleic acid and protein.
  1. For nucleic acid analysis, the spectrophotometer can evaluate samples under various conditions to ascertain the concentration and purity of nucleic acids:
    • 260/280 Ratio
    • Single Ratio
    • Double Ratio
    • ds DNA
    • ss DNA
    • RNA
    • Oligo/DNA Long
    • Oligo/DNA Short
    • Oligo/RNA Long
    • Oligo/RNA Short
  2. In the domain of protein measurement, the device supports several established methods commonly referenced in biochemistry textbooks, including:
    • Direct UV at 280nm
    • Bradford
    • Lowry (Low Sensitivity)
    • Lowry (High Sensitivity)
    • Biuret
    • Colloidal Gold
    • Bicinchoninate (BCA)
  • There are two types of these spectrometers available at this center:
    • DU-800 Single Beam type
    • DU-640 Single Beam type
  • The software allows for measurements in the following categories:
    • Protein
    • Nucleic Acid
    • Kinetics/Time
    • Fixed Wavelength
    • Wavelength Scan
  1. This machine is designed to be used at any time.
  2. The lamp temperature controller ﹝Warm Up﹞time is as follows:
    • VIS → 5 minutes
    • UV → 15 minutes
    • ﹝Reading Kinetics﹞ → 30 minutes
  3. The selection of light sources is as follows:
    • Wavelength below 320nm → UV light source must be turned on
    • Wavelength between 320nm ~ 400nm → UV + VIS must be turned on 
    • Wavelength above 400nm → VIS must be turned on
  4. Use a VIS light source to warm up the machine for 5 minutes; for the UV light source, except for operating ﹝Kinetics/Time﹞, which requires 30 minutes to warm up the machine, other functions only need to warm up the machine for 15 minutes.
    【Due to the high cost of the UV lamp source at around NT$25,000 each and its short lifespan of only 1,000 hours, it is important to use it carefully and efficiently.】
  5. To avoid contamination of the surrounding environment, please don't throw the sample while operating.
  6. Before using the space, please check if the previous user has left any environmental pollution. If so, kindly register it in the registration book to avoid misunderstandings.
  7. Please remember to turn off the light source after use. You don't need to turn off the power of the machine.
