Equipped with three laser light sources with wavelengths of 488nm, 561nm and 633nm.
Details of Three Lasers.
  • To acquire certification for self-operation, you can join the annual summer training course with the BD specialist or watch the training course video on the website. Then, you must be instructed by the BD specialist or certified students before you can request certification from the center.
  • Certification Process
    1. Three hands-on operation practices: For the initial hands-on practice, you should arrange an appointment with the center specialist (Ext:66473 Mr. Chao), and for the second and third times, please schedule an appointment with the seed instructor.
    2. Upon completing the three hands-on practices, you can make an appointment with the center specialist for the certification test. Please make an appointment at least three working days in advance.
    3. This test is essential for obtaining the online reservation and self-operation certification.
  • Notes:
    1. You are required to prepare your own samples for the certification process.
    2. The certification process encompasses understanding flow cytometry principles, pre-startup preparations, standard startup procedures, analyzing samples using software, troubleshooting faults and emergencies, shutdown and cleaning procedures, data export, and maintaining records of usage conditions.
    3. There will be a fee for the three hands-on operation practices.

  • Instrument Reservation
    • The machine is open 24 hours a day. Please make online reservations via the Instrument Reservation System before use.
    • The person making the reservation must be the actual user and cannot be substituted for the reservation.
    • Appointments can be made as early as 14 days in advance, and a maximum of 3 time slots can be reserved, each for up to 4 hours.
    • For technician operation services, make an appointment at least three working days in advance. The appointment time for the technician operation service is 09:00-11:30 and 13:30-16:00 on weekdays (12:00-13:00 is not available).
    • If you are more than half an hour late for the reserved time slot, it will be considered a no-show and giving up the reservation.
       
  • Precautions for use
    1. Users must prepare correction magnetic beads (BD FACS Accudrop Beads, BD#661612).
    2. Follow the correct steps for turning the instrument on and off, and do not make any modifications. Use the provided cleaning supplies such as cotton swabs, Rinse Buffer, Clean Buffer, ddH2O, and Detergent.
    3. The management unit will regularly carry out machine cleaning and maintenance.
    4. Users must follow standard procedures to shut it down, refill PBS, and dispose of the waste liquid after using the instrument. Anyone who fails to turn off the device will be billed for the wear and tear expense.
    5. Register the record book with start and end times for management and billing purposes. Before leaving, clean the instrument and bench and take away any personal items and garbage.
    6. If there are any issues with the instrument, stop using it immediately and contact the instrument management personnel.
    7. The Instrument Center monitors the equipment 24/7 and conducts random inspections. Failure to comply with the above points may result in penalties and compensation for damages.
    8. Technician operation reservation management follows the same procedures as other flow cytometers.
    9. If the sample cap is not removed during sample loading, it will cause deformation of the sample tube and will be compensated according to the price.
    10. Users are responsible for preparing the following items: Pipette, Pipette Tips, paper towels, lens cleaning paper, FACS tubes, archive CD, and Accudrop.

  • If you have the following qualifications, you will be charged the same fee as NYCU faculty and students:
    1. Branch school
    2. Joint-appointed faculty 
    3. Affiliated hospitals 
    4. Projects are done in cooperation with and under the university's control.
  • If you have the following qualifications, the fee will be the same as that of a cooperative educational unit:
    1. Adjunct Faculty
    2. Teaching hospitals (Veterans General Hospital, Taipei City Hospital, Far Eastern Memorial Hospital, etc.)
    3. University System of Taiwan (UST)
    4. Companies participate in the academia-industry cooperation center of NYCU

Users can use different protocols according to their needs, such as antibody labeling or fluorescent gene transfection, but the following procedures must be completed before using the machine:
  1. Samples must be filtered with a nylon mesh (mesh size: 45μm). It is recommended to use Falcon cell strainer #352235 tube with mesh or #352340 40μm mesh. Then, place the filtered cells in a Falcon 12 × 75mm tube (Cat#352052). The table above describes the cell concentration; the minimum sorting volume is 1 mL. Note: Failure to filter the sample, resulting in nozzle blockage or machine damage, will result in liability for compensation. If the machine is clogged and cannot operate due to unfiltered cells, the center reserves the right to cancel the current appointment and charge usage fees.
  2. If the cells need to be cultured after sorting, please prepare a collection tube containing a suitable cell culture medium and hand it over to the operator before the sorting service.
  3. Prepare unstained cells as control group samples for instrument calibration before sorting.
  4. If you sort with multiple fluorescent markers staining, you must provide single stained samples of each fluorescent marker for instrument adjustment and correction before sorting.

  1. No USB flash drive is allowed. Please use FTP to transfer files or bring your blank CD-ROM to burn data. Due to the limited space of the computer's hard disk, the administrator will delete the data from time to time to facilitate the regular operation of the instrument. We are not responsible for data storage.
  2. Without the consent of the administrator of the Center, you are not allowed to add or remove any parts, alter the system settings, or install software on the Center's public equipment. If any experiment requires any test, the administrator's consent must be sought first and accompanied by the engineer of the original manufacturer to carry out the testing without damaging the regular operation of the equipment, and only after the passing of the test can it be operated and used by the personnel approved by the administrator of the Center under the condition of the administrator's permission.
  3. According to the administrator's judgment, the violator is warned for the first time, canceled the right to use for one month for the second time, and canceled the right to use permanently for the third time; if the violator violates the above specification, resulting in the damage of the instrument, the violator shall pay compensation to repair the instrument and stop the right to use for one month. The violator is prohibited from using the instrument permanently.
  • If you have not used the machine for three consecutive months after obtaining the user license, please contact the administrator or the seed instructor to accompany you when operating the machine.
  • Only certified users are allowed to operate the machine. If your lab colleagues need to use the machine, please ask a certified user or reserve commissioned service of the center’s technician to operate the machine on your behalf.
  • The fee for self-operation is NTD300 per half hour for NYCU students and faculty and NTD400 per half hour for technician operation with an additional start-up fee of NTD1,000 (please refer to the rules to apply for the commissioned operation service). The center will calculate the usage hours every two months and bill the laboratories.
  • The instrument computers are for operational use only and cannot be used for other purposes.
    The center and the instrument computer are not responsible for data storage, so please export your data immediately after use.
  • If you encounter any problems during operation, please stop and contact the Seed Instructor Group.
  • After use, please register the usage record (if you have any problems or questions, please note them clearly) to protect your and your laboratory's rights.
  • Unless it is confirmed by the center staff or the original manufacturer that the equipment causes the operation failure, every operation will be charged.
