CytoFLEX A machine is equipped with three laser excitation wavelengths of 488nm (blue), 633nm (red), and 405nm (violet).
It can detect 13 kinds of fluorescent signals at the same time. The detection range and related dyes are as follows:

CytoFLEX B and C machines are equipped with four laser excitation wavelengths: 488nm (blue), 633nm (red), 405nm (violet), and 561nm (yellow).

  • Detection wavelength
    • The device captures scattered light (FSC, SSC) and fluorescent signals (FL1~FL10), along with H, A, and Time as data acquisition parameters. It is capable of detecting particles as small as 200 nm.
    • Absolute counts can be conducted without the need for microspheres.
    • It features a plate autosampler compatible with U, V, and flat bottom 96-well plates. (B & C machine)
    • The system runs on a Windows 7 interface and allows direct graphic exporting to Office-related software without a software lock, facilitating ease of installation on computers.
    • The user interface is designed for simplicity and intuitiveness, enabling adjustments to voltage values and fluorescence compensation via a GUI tool.
    • Boasts a 7 logarithmic ultra-wide dynamic range for data display.
    • Includes a comprehensive matrix fluorescence compensation system, allowing for inter-parameter fluorescence compensation. Compensation experiments can leverage various sample types (e.g., fluorescent beads, cells) for the automatic determination of accurate fluorescence compensation values.
    • Changes to the Gain setting automatically update the fluorescence compensation values, eliminating the manual adjustment requirement.
    • Utilizes the FCS3.0 data storage format for the correction of off-line fluorescence compensation values.
To obtain certification for the CytoFLEX machine, you need to follow these steps:
  1. Download "CytoFLEX Manual (A machine) or CytoFLEX Manual (B&C machine)" and watch the related instructional video (links are at the end of this webpage).
  2. Practice operating the machine under the supervision of certified students.
  3. Reserve a time slot for the certification exam at the center.
  4. The certification exam consists of written and operational tests. If you fail the written test, you won't be able to take the operational test.
    • For the written exam, bring your cell phone to the Center's office during office hours. (Office: 4th floor, Biomedical Engineering Building, extension 62382 or Room B03, Shou-ren Building, extension 66473)
    • For the operational exam, make an appointment at least three working days in advance.
  5. If you're 5 minutes late for the examination, you'll be considered to have given up and failed the examination. If you fail the examination twice, you'll be charged for the third time.
  6. The exam aims to ensure that students can operate the equipment correctly to avoid damage.
  7. The exam includes start-up and shut-down procedures, software operation, and understanding of the instrument's principle. You'll also need to bring samples for the exam.
  8. After receiving the qualified training, you can make an appointment 24 hours before using the "Instrument Reservation System."
Reservation Regulations
  • Make online reservations through the "Instrument Reservation System."
  • The person making the reservation must be the actual user and cannot be substituted.
  • Reservations can be made up to 2 weeks in advance, with a maximum of 4 sessions lasting up to 2 hours each. The maximum number of consecutive appointments for the same laboratory is 4 hours, and 2 hours are required for further appointments.
  • Inform the center in advance of any special experimental needs.
  • If you are late for more than half an hour of the reserved time, it will be considered a violation, and further violations may result in the cancellation of equipment used for one week.
Some precautions for using the machine include:
  • Obtain the certification before use
  • Samples must be filtered by the recommended mesh before loading them onto the device!
  • Use the recommended Spectrum™ nylon mesh (42μM) to filter the sample. It’s recommended to use Falcon cell strainer tube #352235 or 40um mesh #352340.
  • The person who prepares the sample will be responsible for any clogging or damage to the machine caused by loading the sample without filtering.
  • If the sample cap is not removed during sample loading, it will cause deformation of the sample tube and will be compensated according to the price.
  • Maintain the cell concentration of the sample below 106 Events/mL
  • Follow the standard cleaning procedures whenever turning on and off the machine
  • Please record the correct usage time
  • Keep the bench clean after experiments.
  • You will be charged for use if you forget to turn off the machine.
The Instrument Center monitors the equipment 24/7 and conducts random inspections. Failure to comply with the guidelines may result in fines and liability for damages.

Charging standards
  • If you have the following qualifications, you will be charged the same fee as NYCU faculty and students:
    1. Branch school
    2. Joint-appointed faculty 
    3. Affiliated hospitals 
    4. Projects are done in cooperation with and under the control of the university.
  • If you have the following qualifications, the fee will be the same as that of a cooperative educational unit:
    1. Adjunct Faculty
    2. Teaching hospitals (Veterans General Hospital, Taipei City Hospital, Far Eastern Memorial Hospital, etc.)
    3. University System of Taiwan (UST)
    4. Companies participate in the academia-industry cooperation center of NYCU

  1. No USB flash drive is allowed. Please use FTP to transfer files or bring your blank CD-ROM to burn data. Due to the limited space of the computer's hard disk, the administrator will delete the data from time to time to facilitate the normal operation of the instrument. We are not responsible for data storage.
  2. Without the consent of the administrator of the Center, you are not allowed to add or remove any parts, alter the system settings, or install software on the Center's public equipment. If any experiment requires any test, the administrator's consent must be sought first and accompanied by the engineer of the original manufacturer to carry out the testing without damaging the normal operation of the equipment, and only after the passing of the test can it be operated and used by the personnel approved by the administrator of the Center under the condition of the administrator's permission.
  3. According to the administrator's judgment, the violator is warned for the first time, canceled the right to use for one month for the second time, and canceled the right to use permanently for the third time; if the violator violates the above specification, resulting in the damage of the instrument, the violator shall pay compensation to repair the instrument and stop the right to use for one month. The violator is prohibited from using the instrument permanently.
